A transfer membrane receives biomolecules after electrophoresis or another separation step and immobilises them on a surface for detection. Its purpose is often strong target binding, unlike liquid filtration media selected for low adsorption. PVDF and nitrocellulose differ in wetting, handling and detection background, so selection should reflect the target, transfer method and imaging chemistry.
| Selection factor | What to compare |
|---|---|
| Material | PVDF and NC have different preparation and binding characteristics; filtration-grade PVDF is not automatically transfer-grade. |
| Pore structure | Small targets may pass through more readily, making membrane grade and transfer conditions interdependent. |
| Detection | Fluorescence and chemiluminescence place different demands on background and blocking chemistry. |

Align gel and membrane correctly, ensuring even contact without bubbles.
Keep blocking, washing and exposure consistent to separate transfer faults from antibody background.
Nonspecific background, stripping requirements and target detectability also matter.
Investigate bubbles, contact, orientation and drying before changing the transfer parameters.
